    We recently experienced a boy with congenital microcephaly, pre- and post- growth failure, immunodeficiency, bone marrow failure. This patient had a p53 mutation, so that the function of p53 protein was enhanced and the specific symptoms were expected to occur. In humans p53 hyperactive mutation has not been reported as far as we could investigate. We created mice with a p53 mutation equivalent to p53 hyperactivity mutation (p53CTD) due to C-terminal deletion found in patients with advanced bone marrow dysfunction by genetic editing technique. p53CTD / WT mice showed a decrease in peripheral lymphocytes and macrocytic anemia in addition to hyperpigmentation of limbs which is a feature of p53 abnormal activation

    Previously, we performed microarray analysis of etv2 deficient zebrafish embryos, which was one of the earliest markers for angioblasts. We first identified 165 genes that were up-regulated more than 1.5 fold by silencing etv2 expression, then analyzed their expression patterns at 12-somite stage to evaluate whether these genes were associated with the cranial vascular formation, and finally selected 11 genes for the loss of function assay. In this study, we knocked out these 11 genes using the CRISPR/ Cas9 system to analyze the mechanisms of cerebral vasculature formation. As a result, we succeeded in producing sgRNAs for each gene which was capable of the genome editing. For 10 genes, we established heterozygous zebrafish lines in which targeted genes were knocked out by the frame-shift and analysis of the phenotype induced by loss of function were in progress. Additionally, we succeeded in revealing the primary ocular vasculature formation both in zebrafish and mouse embryos.

    This study aimed to develop new genome-editing techniques using engineered nucleases such as CRISPR/Cas9 system in zebrafish. CRISPR/Cas9 system causes DNA double strand breaks, which can be repaired by non-homologous end joining or homology directed recombination. We achieved to knock-in exogenous genes into target genomic locus using donor vector harboring short homologous sequences. The donor vector was designed to have gRNA-recognition sites, homology arms, and the exogenous gene to be integrated. By introducing two gRNA-recognition sites in both side of the integrating gene, backbone sequences of donor vector can be eliminated. Furthermore, the precise genome modification was also occurred in germ cells, resulting in the establishment of knocked-in zebrafish line. This method enables efficient and precise genome modification in a homology-dependent manner, which should be able to be applied to other model organisms

    私は、心臓前駆細胞の移動異常により二股心臓の表現型を示すゼブラフィッシュ変異体の機能解析から、原因遺伝子であるSpns2分子がスフィンゴシン-1-リン酸(S1P)の輸送体として機能することを明らかとした。本研究では、初期発生におけるS1Pの生理機能の全貌を明らかにすることを目的としている。平成22年度は、以下の研究成果が得られた。
    1. ゼブラフィッシュのS1P合成酵素(SphK1,SphK2)およびS1P受容体(S1PR1-S1PR5)の単離を行った。初期発生における発現パターンを解析した結果、S1P合成酵素は、母性遺伝子として供給されており原腸胚期まで胚全体に発現していることが明らかとなった。これに対して、S1P受容体(S1PR1-S1PR5)は、それぞれ特徴的な発現パターンを示しており、様々な器官形成に関与していることが示唆された。
    2. S1Pシグナルと接着分子であるフィブロネクチン(Fn)との遺伝学的な関連性を解析した。Spns2変異体とFn変異体の2重変異体を作製した。Fn変異体は、心臓の形態異常を示すが、正中線への移動異常はほとんど示さない。Spns2変異体の心臓前駆細胞の移動異常は、Fnとの2重変異体とすることによりその移動異常が増強された。このことは、S1Pシグナルとフィブロネクチンが相互に心臓前駆細胞の移動や心臓の形態形成を制御していることを示唆している。
    3. Spns2発現抑制胚、S1PR2発現抑制胚およびコントロール胚からRNAを調整し、マイクロアレイ解析を行った。その結果、34遺伝子の発現の挙動がS1Pシグナル異常で大きく変動していた。脂質代謝に関与する遺伝子(アポリポタンパク質やホスホリパーゼC等)も含まれているので、今後、定量PCR法を用いSpns2変異体での発現を調べ、同定された候補遺伝子は、発現抑制実験を行うことによりS1Pシグナルとの関連性を明らかにする。

    I aimed to reveal novel key molecules in vascular and cardiac development by the characterization of novel cardiovascular zebrafish mutants. In the ko095 mutants, aberrant branching of intersegmental vessels besides of the neural tube was observed. Further, the increased expression of VEGF (vascular endothelial growth factor), which is a key factor in angiogenesis, caused the excessive vascular remodeling in the ko095 mutant. I found that the gene responsible for the ko095 is Sars (Seryl-tRNA synthetase) that is known as an enzyme essential for protein synthesis: Sars possesses a novel function involved in angiogenesis. On the other hand, the ko157 mutant exhibited the impairment of cardiac progenitor migration, resulting in two hearts known as cardia bifida. I found that the gene responsible for the ko157 is a twelve-pass transmembrane-domain protein Spns2 (Spnster-like 2). Biochemical analysis of Spns2 presented that Spns2 functions as a transporter of a sphingolipid mediator, sphingosine-1-phosphate (S1P). Thus, I found that Sars and Spns2 play important roles in vascular and cardiac development in zebrafish, respectively.

    We have performed in situ-based screening to identify genes that are specifically expressed in the blood island where both blood and endothelial cells are generated. In this process, we have isolated the heme synthesis enzymes and further examined the function of CPO (coproporphyrinogen oxidase) during zebrafish erythrogenesis. Knockdown analysis of CPO using anti-sense morpholinos (CPO-MO) caused the strong suppression of hemoglobin production, and the maturation of erythorocyte is morphologically arrested. These results suggest that CPO plays important roles in zebrafish erythrogenesis. Next, we examined the regulation of CPO expression. In zebrafish, both GATA1 and biklf transcriptional factors are essential for the blood cell maturation. Interestingly, CPO promoter region has DNA binding sites for both factors. We found that CPO expression is strongly suppressed when biklf-morpholino was injected into vlt mutant embryos (lacking functional GATA1 gene). In contrast, CPO is ectopically induced when both GATA1 and biklf RNAs are injected into two-cell stage embryos. These results suggest that GATA1 and biklf cooperatively contribute the regulation of CPO expression.
    We have additionally isolated a novel gene, b819 gene that is restrictedly expressed in the blood island. This gene has no homology to any known protein, but primary structure presents a possible membrane protein. Knockdown analysis of b819 gene induced the suppression of blood cells production and the disorganization of network formation of blood vessels. Since both blood and endothelial cells are produced from the common precursor "hemangioblast", b819 gene may be a critical regulator in this process. Further studies will be required to reveal how b819 gene contributes the development of blood and endothelial cells during early embryogenesis.

  • Functional analysis of a novel gene arf in the brain development during early zebrafish embryogenesis

    We have isolated a novel factor arf (anterior brain regulating factor) that is specifically expressed in the anterior brain during early zebrafish embryogenesis. In this study, we have performed the functional analysis of arf. At first, we examined the expression pattern of arf by the whole-mount in situ hybridization analysis. (1) We found that arf is expressed in the anterior ectoderm (future anterior brain) during gastrulation stages and expressed in the anterior brain (forebrain and midbrain) and the formed somites during somitogenesis stages. Next, we investigated the knockdown analysis of the arf using anti-sense morpholinos for the arf gene (arf-MO). (2) Injection of arf-MO in the blastomere of the one-cell stage embryo induced the disorganization of anterior brain and the curly tail defects. These abnormalities are well correlated with the arf expression domains. These results suggest that arf plays an important role in the formation of brain and trunk including somites. We examined whether apoptotic cell death is affected in the arf-MO-injected embryos. (3) TUNEL assay showed that apoptotic cell death is broadly induced including both brain and trunk in the arf-MO-injected embryos. We finally examined the effect of arf overexpression. (4)We could not observe any morphological effect in the arf RNA-injected embryos. We should check the dose of arf RNA and the effect on the marker genes expression. These results suggest that arf is involved in the brain and trunk formation. Further studies will be required to elucidate how arf contributes the formation of anterior brain in early embryogenesis.
