    Ecological divergence in a species provides a valuable opportunity to study the early stages of speciation. We focused on <italic>Metrosideros polymorpha</italic>, a unique example of the incipient radiation of woody species, to examine how an ecological divergence continues in the face of gene flow. We analyzed the whole genomes of 70 plants collected throughout the island of Hawaii, which is the youngest island with the highest altitude in the archipelago and encompasses a wide range of environments. The continuous <italic>M</italic>. <italic>polymorpha</italic> forest stands on the island of Hawaii were differentiated into three genetic clusters, each of which grows in a distinctive environment and includes substantial genetic and phenotypic diversity. The three genetic clusters showed signatures of selection in genomic regions encompassing genes relevant to environmental adaptations, including genes associated with light utilization, oxidative stress, and leaf senescence, which are likely associated with the ecological differentiation of the species. Our demographic modeling suggested that the glaberrima cluster in wet environments maintained a relatively large population size and two clusters split: polymorpha in the subalpine zone and incana in dry and hot conditions. This ecological divergence possibly began before the species colonized the island of Hawaii. Interestingly, the three clusters recovered genetic connectivity coincidentally with a recent population bottleneck, in line with the weak reproductive isolation observed in the species. This study highlights that the degree of genetic differentiation between ecologically-diverged populations can vary depending on the strength of natural selection in the very early phases of speciation.

    Key message The roles of root exudates are not to directly solubilize P bound to non-crystalline minerals but to increase organic-P and plant-available P through priming effects for microbes on volcanic soils. Volcanic soils are generally characterized by low phosphorus (P) availability for plants because the non-crystalline minerals produced during weathering process strongly combine P with ligand binding. How forest trees absorb such bound P in volcanic soils have not been well known. The aim of this study was to investigate the mechanisms of P acquisition by trees in three evergreen broadleaved forests on volcanic soils differing in the concentration of non-crystalline minerals with an emphasis on the role of low-molecular-weight organic acids (LMWOAs). LMWOAs released from tree fine roots were expected (1) to chemically release P bound to non-crystalline minerals, and/or (2) to biologically enhance P availability through priming effects by stimulating soil microbial activities and by increasing the concentration of organic P in the rhizosphere. Compositions of LMWOA exudates were phylogenetically determined but the flux of citric and malic acids tended to increase in soils where the concentration of non-crystalline minerals was higher. The concentration of P bound to non-crystalline minerals was lower in the rhizosphere than in non-rhizosphere soils, suggesting the solubilization of the bound P in the rhizosphere; however, this trend occurred only in one forest. In contrast, the concentrations of organic P, easily soluble P and total P were significantly greater in the rhizosphere than in non-rhizosphere soils in all sites. Our results suggested that LMWOAs exudates did not directly release P from non-crystalline minerals but increased organic P associated with soil microbial biomass in rhizosphere soils, which in turn enhanced plant-available P and total P.

    <title>Abstract</title>Improved phosphorus (P) use efficiency for crop production is needed, given the depletion of phosphorus ore deposits, and increasing ecological concerns about its excessive use. Root system architecture (RSA) is important in efficiently capturing immobile P in soils, while agronomically, localized P application near the roots is a potential approach to address this issue. However, the interaction between genetic traits of RSA and localized P application has been little understood. Near-isogenic lines (NILs) and their parent of rice (<italic>qsor1</italic>-NIL, <italic>Dro1</italic>-NIL, and IR64, with shallow, deep, and intermediate root growth angles (RGA), respectively) were grown in flooded pots after placing P near the roots at transplanting (P-dipping). The experiment identified that the P-dipping created an available P hotspot at the plant base of the soil surface layer where the <italic>qsor1</italic>-NIL had the greatest root biomass and root surface area despite no genotyipic differences in total values, whereby the <italic>qsor1</italic>-NIL had significantly greater biomass and P uptake than the other genotypes in the P-dipping. The superior surface root development of <italic>qsor1</italic>-NIL could have facilitated P uptakes from the P hotspot, implying that P-use efficiency in crop production can be further increased by combining genetic traits of RSA and localized P application.

    We measured the productivity and morphological traits of fine roots in seven forests between 170 and 1550 m along an elevation gradient on Yakushima Island, Japan. Yakushima Island is continuously covered by forests ranging from the lowland evergreen broad-leaved forests below 1000 m to the upland evergreen coniferous forests above. Mean annual temperature decreases linearly and soil nutrient availabilities (both N and P) also decrease with increasing elevation. We employed an in-growth core method to determine fine-root productivity. Mean fine-root biomass ranged from 0.123 to 0.297 kg m −2 in the lowland broad-leaved forests and from 0.092 to 0.097 kg m −2 in the upland coniferous forests. Mean fine-root productivity ranged from 0.063 to 0.173 kg m −2  yr −1 in the lowland broad-leaved forests and from 0.033 to 0.044 kg m −2  yr −1 in the upland coniferous forests. Both mean fine-root biomass and mean fine-root productivity were significantly greater in middle-elevation broad-leaved forests than in the lower broad-leaved or the higher coniferous forests. Fine-root turnover rate tended to be lower and mean specific root length (SRL) tended to be higher in the coniferous forests. In the upland coniferous forests, trees may adapt to nutrient limitation and/or cooler climate by increasing the life span and SRL of fine roots instead of increasing carbon allocation to fine roots, thereby decreasing fine-root productivity. Fine-root dynamics along the elevational gradient on Yakushima reflected the species shift from broad-leaved to coniferous trees rather than the monotonous reduction of temperature and nutrient availability.
