    Duckweeds (Lemnaceae) are representative producers in fresh aquatic ecosystems and also yield sustainable biomass for animal feeds, human foods, and biofuels, and contribute toward effective wastewater treatment; thus, enhancing duckweed productivity is a critical challenge. Plant-growth -promoting bacteria (PGPB) can improve the productivity of terrestrial plants; however, duckweed-PGPB interactions remain unclear and no previous study has investigated the molecular mechanisms underlying duckweed-PGPB interaction. Herein, a PGPB, Ensifer sp. strain SP4, was newly isolated from giant duckweed (Spirodela polyrhiza), and the interactions between S. polyrhiza and SP4 were investigated through physiological, biochemical, and metabolomic analyses. In S. polyrhiza and SP4 coculture, SP4 increased the nitrogen (N), chlorophyll, and ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO) contents and the photosynthesis rate of S. polyrhiza by 2.5-, 2.5-, 2.7-, and 2.4-fold, respectively. Elevated photosynthesis increased the relative growth rate and biomass productivity of S. polyrhiza by 1.5 and 2.7-fold, respectively. Strain SP4 significantly altered the metabolomic profile of S. polyrhiza, especially its amino acid profile. N stable isotope analysis revealed that organic N compounds were transferred from SP4 to S. polyrhiza. These N compounds, particularly glutamic acid, possibly triggered the increase in photosynthetic and growth activities. Accordingly, we propose a new model for the molecular mechanism underlying S. polyrhiza growth promotion by its associated bacteria Ensifer sp. SP4, which occurs through enhanced N compound metabolism and photosynthesis. Our findings show that Ensifer sp. SP4 is a promising PGPB for increasing biomass yield, wastewater purification activity, and CO2 capture of S. polyrhiza.

    The abundance and diversity of anaerobic ammonium oxidation (anammox) bacteria were assessed in 152 groundwater samples in the Kathmandu Valley, Nepal. Anammox bacterial 16S rRNA genes were detected in 54% (37/68) of samples collected in the dry season at 1.6×105-8.8×106 copies L-1, and in 60% (50/84) of samples collected in the wet season at 4.3×104-1.2×107 copies L-1. The 16S rRNA genes of "Candidatus Brocadia", "Candidatus Anammoxoglobus", and five new deduced anammox bacterial phylotypes were detected in the shallow groundwater samples. Diverse anammox bacteria were broadly distributed in the shallow groundwater aquifer of the Kathmandu Valley.

    The microbial communities inhabiting the fronds of duckweeds have not been investigated in as much detail as those on the roots. We herein examined the microbial communities in three duckweed species using 16S rRNA amplicon sequencing and compared them to those on the roots. The microbial compositions of the fronds were distinct from those of the roots in the three species. Various types of taxonomic bacteria, including rarely cultivated phyla, Acidobacteria, Armatimonadetes, and Verrucomicrobia, were also isolated from the fronds, but at a slightly lower abundance than those from the roots. These results suggest that duckweed fronds are an alternative source for isolating rare and novel microbes, which may otherwise be recalcitrant to cultivation using conventional strategies.

    A dropping nitrification reactor was proposed as a low-cost and energy-saving option for the removal of NH4+-N from contaminated groundwater. The objectives of this study were to investigate NH4+-N removal performance and the nitrogen removal pathway and to characterize the microbial communities in the reactor. Polyolefin sponge cubes (10 mm x 10 mm x 10 mm) were connected diagonally in a nylon thread to produce 1 m long dropping nitrification units. Synthetic groundwater containing 50 mg L-1 NH4+-N was added from the top of the hanging units at a flow rate of 4.32 L day(-1) for 56 days. Nitrogen-oxidizing microorganisms in the reactor removed 50.8-68.7% of the NH4+-N in the groundwater, which was aerated with atmospheric oxygen as it flowed downwards through the sponge units. Nitrogen transformation and the functional bacteria contributing to it were stratified in the sponge units. Nitrosomonadales-like AOB predominated and transformed NH4+-N to NO2--N in the upper part of the reactor. Nitrospirales-like NOB predominated and transformed NO2--N to NO3--N in the lower part of the reactor. The dropping nitrification reactor could be a promising technology for oxidizing NH4+-N in groundwater and other similar contaminated wastewaters.

    © 2019 Elsevier Ltd Plant growth-promoting bacteria (PGPB) are considered a promising tool to improve biomass production and water remediation by the aquatic plant, duckweed; however, no effective methodology is available to utilize PGPB in large hydroponic systems. In this study, we proposed a two-step cultivation process, which comprised of a “colonization step” and a “mass cultivation step,” and examined its efficacy in both bucket-scale and flask-scale cultivation experiments. We showed that in the outdoor bucket-scale experiments using three kinds of environmental water, plants cultured through the two-step cultivation method with the PGPB strain, Acinetobacter calcoaceticus P23, yielded 1.9 to 2.3 times more biomass than the control (without PGPB inoculation). The greater nitrogen and phosphorus removals compared to control were also attained, indicating that this strategy is useful for accelerating nutrient removal by duckweed. Flask-scale experiments using non-sterile pond water revealed that inoculation of strain P23 altered duckweed surface microbial community structures, and the beneficial effects of the inoculated strain P23 could last for 5–10 d. The loss of the duckweed growth-promoting effect was noticeable when the colonization of strain P23 decreased in the plant. These observations suggest that the stable colonization of the plant with PGPB is the key for maintaining the accelerated duckweed growth and nutrient removal in this cultivation method. Overall, our results suggest the possibility of an improved duckweed production using a two-step cultivation process with PGPB.

    Background: Euglena gracilis, a unicellular flagellated microalga, is regarded as one of the most promising species as microalgal feedstock for biofuels. Its lipids (mainly wax esters) are suitable for biodiesel and jet fuel. Culture of E. gracilis using wastewater effluent will improve the economics of E. gracilis biofuel production. Enhancement of the productivity of E. gracilis biomass is critical to creating a highly efficient biofuels production system. Certain bacteria have been found to promote microalgal growth by creating a favorable microenvironment. These bacteria have been characterized as microalgae growth-promoting bacteria (MGPB). Co-culture of microalgae with MGPB might offer an effective strategy to enhance microalgal biomass production in wastewater effluent culture systems. However, no MGPB has been identified to enhance the growth of E. gracilis. The objectives of this study were, therefore, to isolate and characterize the MGPB effective for E. gracilis and to demonstrate that the isolated MGPB indeed enhances the production of biomass and lipids by E. gracilis in wastewater effluent culture system. Results: A bacterium, Emticicia sp. EG3, which is capable of promoting the growth of microalga E. gracilis, was isolated from an E. gracilis-municipal wastewater effluent culture. Biomass production rate of E. gracilis was enhanced 3.5-fold and 3.1-fold by EG3 in the co-culture system using a medium of heat-sterilized and non-sterilized wastewater effluent, respectively, compared to growth in the same effluent culture but without EG3. Two-step culture system was examined as follows: E. gracilis was cultured with or without EG3 in wastewater effluent in the first step and was further grown in wastewater effluent in the second step. Production yields of biomass and lipids by E. gracilis were enhanced 3.2-fold and 2.9-fold, respectively, in the second step of the system in which E. gracilis was co-cultured with EG3 in the first step. Conclusion: Emticicia sp. EG3 is the first MGPB for E. gracilis. Growth-promoting bacteria such as EG3 will be promising agents for enhancing E. gracilis biomass/biofuel productivities.

    Rationale: The isotopic tracer technique is widely used to identify the sources and fate of nitrogen (N) in order to understand the N cycle and contamination in water environments. The stable isotope ratio of ammonium is expected to greatly enhance the tracing analysis by combining it with the traditional nitrate isotope ratio. Diffusion followed by gas-phase trapping is the most commonly applied method for ammonium isotope ratio measurement. Although dissolved organic nitrogen (DON) is abundant in natural water and its breakdown in the diffusion procedure has been reported, the interference of DON with the measurement of ammonium isotope ratios has not been fully examined. Methods: This study aims to test the effect of DON contamination by using organic N compounds, viz. humic acid and alanine. A series of diffusion experiments was conducted at a temperature of 80°C for a maximum of 7 days. Ammonia was transferred from alkaline solution and trapped with an acidic filter. This method was applied for samples with ammonium concentrations between 0.5 and 2.0 mg-N/L. Results: There was no difference between the ammonium N stable isotope ratios for samples with and without added DON compounds
    the fractionation between the observed value and the actual value was negligible, in the range of 0.2 to 1.0‰. The modifications of previous studies, i.e. shorter diffusion period, no vigorous shaking and using gas-phase trapping, successfully avoided any breakdown of DON in fresh water samples. Conclusions: The modified method provides high precision and accuracy and it is recommended for the analysis of anthropogenically influenced water samples, such as paddy fields, ground water, rivers and lakes.

    To assess the potential of duckweeds as agents for nitrogen removal and biofuel feedstocks, Spirodela polyrhiza, Lemna minor, Lemna gibba, and Landoltia punctata were cultured in effluents of municipal wastewater, swine wastewater, or anaerobic digestion for 4 days. Total dissolved inorganic nitrogen (T-DIN) of 20–50 mg/L in effluents was effectively removed by inoculating with 0.3–1.0 g/L duckweeds. S. polyrhiza showed the highest nitrogen removal (2.0–10.8 mg T-DIN/L/day) and biomass production (52.6–70.3 mg d.w./L/day) rates in all the three effluents. Ethanol and methane were produced from duckweed biomass grown in each effluent. S. polyrhiza and L. punctata biomass showed higher ethanol (0.168–0.191, 0.166–0.172 and 0.174–0.191 g-ethanol/g-biomass, respectively) and methane (340–413 and 343–408 NL CH4/kg VS, respectively) production potentials than the others, which is related to their higher carbon and starch contents and calorific values.

    Duckweed offers the promise of a co-benefit culture combining water purification with biomass production. Acinetobacter calcoaceticus P23 is a plant growth-promoting bacterium isolated from a duckweed, Lemna aequinoctialis. This study quantified its growth-promoting effect on three duckweeds (L. aoukikusa, L. minor, and Spirodela polyrhiza) in sterile Hoagland solution and evaluated its usefulness in duckweed culture under non-sterile conditions. P23 promoted growth of three duckweeds in sterile Hoagland solution at low to high nutrient concentrations (1.25-10 mg NO3-N/L and 0.25-2.0 mg PO4-P/L). It increased the biomass production of L. aequinoctialis 3.8-4.3-fold, of L. minor 2.3-3.3-fold, and of S. polyrhiza 1.4-1.5-fold after 7 days compared with noninoculated controls. P23 also increased the biomass production of L. minor 2.4-fold in pond water and 1.7-fold in secondary effluent of a sewage treatment plant under non-sterile conditions at laboratory-scale experiments. P23 rescued L. minor from growth inhibition caused by microorganisms indigenous to the pond water. The results demonstrate that the use of P23 in duckweed culture can improve the efficiency of duckweed biomass production, and a positive effect of P23 on duckweed-based wastewater treatment can be assumed.

    A flask-scale laboratory study was performed to assess the bioaugmentation of water contaminated with a mixture of 2-nitrophenol, 3-nitrophenol, 4-nitrophenol and 2,4-dinitrophenol by using a bacteria consortium consisting of three nitrophenol-degrading bacteria strains (Pseudomonas sp. strain MFR-1, Pseudomonas sp. strain PFR-1 and Rhodococcus sp. strain DFR-1), reinoculated into the roots of Spirodela polyrhiza. The selected strains were colonized into the root at approximately 10(4) to 10(6) colony-forming units (CFU per plant). The high populations remained stable through five sequential two-days degradation cycles and complete nitrophenol removal was achieved within five-repeated cycles. Hence, inoculation of subjected degraders into the roots of aquatic plants is an effective treatment for nitrophenol-contaminated effluents or aquatic resources.

    We investigated the use of Novosphingobium sp. strain TYA-1 for the simultaneous removal of bisphenol A (BPA) and 4-alkylphenols (4-APs) from complex polluted waters. Strain TYA-1 degraded BPA and utilized it as a sole carbon and energy source via oxidative skeletal rearrangement involving the cytochrome p450 monooxygenase system. Strain TYA-1 also degraded 4-APs with branched side alkyl chains (4-tert-butylphenol [4-tert-BP], 4-sec-butylphenol, 4-tert-pentylphenol, 4-tert-octylphenol [4-tert-OP], and branched nonylphenol mixture) via 4-alkylcatechols but could not degrade 4-APs with linear side alkyl chains. Degradation of 4-APs, like that of BPA, involved the cytochrome p450 monooxygenase system in strain TYA-1. A sequencing batch bioreactor (100 mL of polluted water [50 mg/L BPA, 50 mg/L 4-tert-BP, and 5 mg/L 4-tert-OP]; 6 h of reaction time/cycle; 12 cycles in total) containing alginate-immobilized TYA-1 cells (15 mg dry cells) simultaneously removed BPA, 4-tert-BP, and 4-tert-OP from complex polluted waters. These immobilized TYA-1 cells could be reused for a total of 9 cycles without any loss of degradation activity. Our results support the potential of using immobilized TYA-1 cells for the simultaneous removal of BPA and 4-APs from complex polluted waters.

    We investigated the biodegradation of 2-nitrophenol (2-NP), 4-nitrophenol (4-NP), and 2,4-dinitrophenol (2,4-DNP) in the rhizosphere of Spirodela polyrrhiza plants by conducting degradation experiments with three river water samples supplemented with each nitrophenol (NP). We then isolated NP-degrading bacteria both from the S. polyrrhiza roots and from the river water. In the river water samples, removal of the three NP was accelerated in the presence of S. polyrrhiza plants. The three NPs persisted in an autoclaved solution with sterile plants suggests that NP removal was accelerated largely by bacterial NP biodegradation rather than by adsorption and uptake by the plants. We isolated 8 strains of NP-degrading bacteria: 6 strains from the S. polyrrhiza roots and 2 strains from river water without the plants. The 2-NP- and 2,4-DNP-degrading bacteria were isolated only from the S. polyrrhiza roots. The 4-NP-degrading bacteria different from those isolated from the river water samples were also found on S. polyrrhiza roots. The 2-NP- and 4-NP-degrading strains isolated from the roots utilized the corresponding NP (0.5 mmol/L) as the sole carbon and energy source. The 2,4-DNP-degrading strains isolated from the roots showed substantial 2,4-DNP-degrading activity, but the presence of other carbon and energy sources was required for their growth. The isolated NP-degrading bacteria from the roots must have contributed to the accelerated degradation of the three NPs in the rhizosphere of S. polyrrhiza. Our results suggested that rhizoremediation with S. polyrrhiza may be effective for NP-contaminated surface water.

    We investigated rhizoremediation using Phragmites australis (common reed) for fresh sediment contaminated with 4-n-butylphenol (4-n-BP). We collected P. australis plant, rhizosphere sediment, and unvegetated sediment samples from a natural pond and conducted degradation experiments using sediments spiked with 4-n-BP. 4-n-BP was rapidly removed from the rhizosphere sediment with P. australis plants, whereas 4-n-BP persisted in the unvegetated sediment without plants and in the autoclaved rhizosphere sediment with P. australis plants, suggesting that the accelerated 4-n-BP removal resulted largely from the biodegradation by rhizosphere bacteria rather than by adsorption and uptake by the plant. Four bacterial strains, Pseudomonas spp. TO-1, TO-2, TO-3, and TO-4, capable of utilizing 4-n-BP as a sole carbon source via a meta-cleavage pathway, were isolated from the rhizosphere sediment. These strains presumably contributed to the accelerated 4-n-BP removal from the rhizosphere sediment. Oxygen and organic compounds from the roots elevated both cell numbers and 4-n-BP-degrading activity of strain TO-4 (the only strain tested) in the rhizosphere sediment, leading to rapid 4-n-BP removal. The results suggest that rhizoremediation using P. australis can be applicable to remove 4-n-BP from the contaminated sediments.

    The bioremediation potential of crude oil by Polyporus sp. S133 pre-grown in wood meal was investigated in two separate experiment trials; liquid medium and soil. The effect of three nutrients (glucose, polypeptone, and wood meal), oxygen flow, and some absorbent on the efficiency of the process was also evaluated. Degradation of crude oil in soil was significantly increased with an addition of oxygen flow and some absorbent (kapok and pulp). The highest degradation rate of crude oil was 93% in the soil with an addition of 10% kapok. The present study clearly demonstrates that, if suitably developed, Polyporus sp. S133 could be used to remediate soil contaminated with crude oil.

    We investigated biodegradation of technical nonylphenol (tNP) in Phragmites australis rhizosphere sediment by conducting degradation experiments using sediments spiked with tNP. Accelerated tNP removal was observed in P. australis rhizosphere sediment, whereas tNP persisted in unvegetated sediment without plants and in autoclaved sediment with sterile plants, suggesting that the accelerated tNP removal resulted largely from tNP biodegradation by rhizosphere bacteria. Three bacterial strains, Stenotrophomonas sp. strain IT-1 and Sphingobium spp. strains IT-4 and IT-5, isolated from the rhizosphere were capable of utilizing tNP and 4-tert-octylphenol as a sole carbon source via type II ipso-substitution. Oxygen from P. australis roots, by creating highly oxygenated conditions in the sediment, stimulated cell growth and the tNP-degrading activity of the three strains. Moreover, organic compounds from P. australis roots functioned as carbon and energy sources for two strains, IT-4 and IT-S, supporting cell growth and tNP-degrading activity. Thus, P. australis roots elevated the cell growth and tNP-degrading activity of the three bacterial strains, leading to accelerated tNP removal. These results demonstrate that rhizoremediation of tNP-contaminated sediments using P. australis, can be an effective strategy.

    We investigated the biodegradation of pyrene and benzo[a]pyrene in Phragmites australis rhizosphere sediment. We collected P. australis plants, rhizosphere sediments, and unvegetated sediments from natural aquatic sites and conducted degradation experiments using sediments spiked with pyrene or benzo[a]pyrene. Accelerated removal of pyrene and benzo[a]pyrene was observed in P. australis rhizosphere sediments with plants, whereas both compounds persisted in unvegetated sediments without plants and in autoclaved rhizosphere sediments with sterilized plants, suggesting that the accelerated removal resulted largely from biodegradation by rhizosphere bacteria. Initial densities of pyrene-utilizing bacteria were substantially higher in the rhizosphere than in unvegetated sediments, but benzo[a]pyrene-utilizing bacteria were not detected in rhizosphere sediments. Mycobacterium gilvum strains isolated from rhizosphere sediments utilized pyrene aerobically as a sole carbon source and were able to degrade benzo[a]pyrene when induced with pyrene. Phragmites australis root exudates containing phenolic compounds supported growth as a carbon source for the one Mycobacterium strain tested, and induced benzo[a] pyrene-degrading activity of the strain. The stimulatory effect on benzo[a]pyrene biodegradation and the amounts of phenolic compounds in root exudates increased when P. australis was exposed to pyrene. Our results show that Mycobacterium-root exudate interactions can accelerate biodegradation of pyrene and benzo[a]pyrene in P. australis rhizosphere sediments. (C) 2010 Elsevier Ltd.     Removal experiments of phenol, aniline, 2,4-dichlorophenol, nonylphenol and bisphenol A (BPA) using Spirodela polyrrhiza-bacterial associations revealed that all compounds but BPA underwent accelerated removal. The mechanisms differed depending on the substrates. It was found that S. polyrrhiza has a great ability to release phenolic compound-rich root exudates, and the exudates seem to stimulate bacterial degradation of a variety of aromatic compounds.

    The bacterial community structure in bulk water and in rhizosphere fractions of giant duckweed, Spirodela polyrrhiza, was quantitatively and qualitatively investigated by PCR-based methods using 6 environmental water samples to elucidate the mechanisms underlying selective accumulation of aromatic compound-degrading bacteria in the rhizosphere of S. polyrrhiza. S. polyrrhiza selectively accumulated a diverse range of aromatic compound-degrading bacteria in its rhizosphere, regardless of the origin of water samples, despite no exposure to phenol. The relative abundances of the catechol 1,2-dioxygenase (C12O) gene (C12O DNA) and catechol 2,3-dioxygenase (C23O) gene (C23O DNA) were calculated as the ratios of the copy numbers of these genes to the copy number of 16S rDNA and are referred to as the rhizosphere effect (RE) value. The RE values for C12O DNA and C23O DNA were 1.0 x 10(1)-9.3 x 10(3) and 1.7 x 10(2)-1.5 x 10(4) times as high, respectively, in rhizosphere fractions as in bulk water fractions, and these higher values were associated with a notably higher sequence diversity of C12O DNA and C23O DNA. The RE values during phenol degradation were 3.6 x 10(0)-4.3 x 10(2) and 2.2 x 10(0)-1.7 x 10(2), respectively, indicating the ability of S. polyrrhiza to selectively accumulate aromatic compound-degrading bacteria in its rhizosphere during phenol degradation. The bacterial communities in the rhizosphere fractions differed from those in the bulk water fractions, and those in the bulk water fractions were notably affected by the rhizosphere bacterial communities. S. polyrrhiza released more than 100 types of phenolic compound into its rhizosphere as root exudates at the considerably high specific release rate of 1520 mg TOC and 214 mg phenolic compounds/d/g root (wet weight). This ability of S. polyrrhiza might result in the selective recruitment and accumulation of a diverse range of bacteria harboring genes encoding C12O and C23O, and the subsequent accelerated degradation of phenol in the rhizosphere.     The accelerated removal of bisphenols A and F (BPA, BPF) was observed in the rhizosphere sediment of Phragmites australis, while they persisted in the absence of P. australis. A BPA-degrading bacterium, Novosphingobium sp. strain TYA-1, and a BPF-degrading bacterium, Sphingobium yanoikuyae strain TYF-1, were isolated from the rhizosphere of P. australis.     Changes of possible key catabolic genes and microbial community structures during the degradation of NPEOs and NP in natural water microcosms were investigated using the most-probable-number-polymerase chain reaction (MPN-PCR) and terminal restriction fragment length polymorphism (T-RFLP). The copy number of catechol 2,3-dioxygenase (C23O) DNA increased significantly during NPEO and NP degradation, suggesting that meta-cleavage of the aromatic rings of NPEOs and NP might have happened. Catechol 1,2-dioxygenase (C12O) DNA, alkane-catabolic genes (alk), and 16S rDNA, on the other hand, did not change notably, suggesting that the two genes might not be the relevant genes for NPEOs and NP degradation. The 16S rRNA gene-based T-RFLP analysis results indicated that specific and different dominant (or degrading) bacteria should be selected, depending on the substances. A strain with a DNA length of 78 bp, which might be affiliated with the beta subclass of Proteobacteria, became the dominant species for NPEO degradation, while strains at 88 and 198 bp were dominant in the NP microcosm. Diversity of microbial community structure tended to be simplified after NPEO degradation, while that in the NP microcosm remained relatively stable. Five clusters were obtained according to the similarity in community structures of different microcosms by cluster analysis, which were consistent with the biodegradation behaviors of different microcosms.     Accelerated degradation of organic chemicals by aquatic plant-bacterial associations was reported for the first time with elucidation of the role and contribution of aquatic plant and bacteria in its rhizosphere using a fast-growing giant duckweed, Spirodela polyrrhiza. The results clearly showed the accelerated degradation of all the three aromatic compounds (phenol, aniline and 2,4-dichlorophenol [2,4-DCP]) tested by aquatic plant-bacterial associations. In phenol degradation system, phenol-degrading bacteria indigenous to the rhizosphere fraction of S. polyrrhiza mainly contributed, while in aniline degradation system S. polyrrhiza mainly contributed by stimulating aniline-degrading bacteria both in the rhizosphere and balk water fraction. On the other hand in 2,4-DCP degradation system, S. polyrrhiza itself mainly contributed to its removal by uptake and degradation. Thus, the mechanisms for accelerated removal of aromatic compounds were quite different depending on the substrates. S. polyrrhiza showed selective accumulation of phenol-degrading bacteria in its rhizosphere fraction, while aniline- and 2,4-DCP-degrading bacteria were not much accumulated. S. polyrrhiza secreted peroxidase and laccase. However, both of the enzymatic activities increased with the addition of aromatic compounds, degrading ability of S. polyrrhiza itself should be owing to the production of peroxidase rather than laccase because the change of peroxidase activity and concentration of each aromatic compound well concurred. From the results obtained in the present study, it can be concluded that the feasibility of the use of aquatic plant-bacterial associations to accelerate the degradation of organic chemicals especially recalcitrant compounds in aquatic environment was shown.
